MEFs can be reprogramed to pluripotent stem cells by defined reprogramming factors1. However, the reprograming efficiency is influenced by kinds of conditions, such as the combination of reprogramming factors, culture medium, types of the virus and the cell density etc2. This protocol described the optimized condition for reprogramming MEFs by different reprogramming system, especially the combination of different reprogramming factors.
